Green one-pot synthesis of pyrano[2,3-c]pyrazole monoazo dyes for high-performance textile dyeing and AgNP-enabled antimicrobial/UV functionalization Pages 691-710 PDF Download PDF

Authors: Ahmed Ragheb, Maher H. Helal, Saadia A. Abd El-Megied, Fatma A. Mohamed, Mohamed G. Abouelenein

doi 10.5267/j.ccl.2026.3.002

๐Ÿ”‘ Keywords: Pyrano[2, 3-c]pyrazole, Synthesis, Monoazo dyes, Silver nanoparticles, Textile, Antibacterial textiles

Abstract:
Despite extensive reports on azo dyes, the development of environmentally benign and multifunctional dye systems tailored for technical textiles remains limited. In this work, a green one-pot multicomponent strategy was employed to synthesize a series of pyrano[2,3-c]pyrazole-based monoazo dyes, which were structurally validated by comprehensive spectroscopic and elemental analyses. The dyes exhibited high exhaustion and fixation on polyester, nylon, and wool fabrics, delivering uniform deep shades with strong color strength, excellent fastness properties, and compatibility with high-temperature dyeing without auxiliary fixation steps. Notably, post-dyeing silver nanoparticle functionalization imparted pronounced and durable antimicrobial activity while preserving chromatic performance.

Indoleโ€“1,2,4-triazole hybrids as selective ERK inhibitors: Synthesis, anticancer evaluation, and molecular modeling Pages 851-866 PDF Download PDF

Authors: Mohamed G. Abouelenein, Yasmin K. Elsersy, Hayam A. Abd El Salam, Hanem M. Awad, Zeinab A. El-Shahid, Mohammed T. Abdel-Aal, Heba M. Abo-Salem

doi 10.5267/j.ccl.2025.7.003

๐Ÿ”‘ Keywords: Indole-3-propionic acid, Anticancer agents, Cytotoxicity, ERK kinase, Molecular docking, ADMET

Abstract:
A new series of structurally diverse indoleโ€“1,2,4-triazole derivatives was designed and synthesized through a thiolated triazole intermediate derived from indole-3-propionic acid. The final compounds, including S-acetamide, bis-indolyl, triazolothiadiazole, and triazolothiadiazine analogues (3โ€“8), were confirmed by NMR, IR, MS, and elemental analysis. Their cytotoxicity was tested against breast (MCF-7), colon (HCT-116), and liver (HepG2) cancer cell lines, alongside normal BJ-1 fibroblasts, using the LDH assay. Several compounds, especially 3, 6b, 7a, 7c, and 8a,b showed potent and selective antiproliferative effects on MCF-7 and HCT-116 cells with IC50 values between 2.3 and 3.0 ฮผM, outperforming doxorubicin nearly twofold under the same conditions. Mechanistic studies revealed a significant decrease in phosphorylated ERK (pERK) protein levels in MCF-7 cells, with compound 3b showing the strongest inhibition (3.499 ยฑ 0.21 pg/mL), consistent with its IC50 (2.3 ฮผg/mL). Molecular docking supported the strong binding affinity of 3b within the ERK active site (โˆ’9.0 kcal/mol), involving hydrogen bonding and hydrophobic interactions. In silico ADMET predictions confirmed favorable drug-likeness, oral bioavailability, and pharmacokinetic safety for this compound. Overall, compound 3b emerges as a promising lead for ERK-targeted anticancer drug development, supported by combined synthetic, biological, and computational evidence.

Synthesis, molecular docking studies, and antimicrobial evaluation of pyrano[2, 3-c]pyrazole derivatives Pages 309-328 PDF Download PDF

Authors: Samy A. El-Assaly, Abd El-Hamid A. Ismail, Hamed Abdel Bary, Mohamed G. Abouelenein

doi 10.5267/j.ccl.2021.3.003

๐Ÿ”‘ Keywords: Pyrano[2, 3-c]pyrazole, Spirooxindoles, Pyrimidine, Antimicrobial, Molecular docking

Abstract:
A sequence of pyrano[2, 3-c]pyrazoles was constructed through promoting an eco-friendly, green, and efficient approach. M1-M25 derivatives were developed by a base-catalyzed one-pot reaction involving application of hydrazine hydrate 96%, ฮฒ-keto ester as ethyl acetoacetate or diethyl malonate, aryl/heteroaryl aldehyde or isatin, and enolizable active methylene compounds with isolation of unexpected compound M2. Further on, intramolecular cyclization of compounds M10, M13 with formic acid, acetic anhydride, and formamide leads to the corresponding pyrimidine derivatives M26-M31. Afterwards, the antimicrobial activity of the compounds was evaluated and fortunately, the vast majority of the compounds showed outstanding anti-bacterial results. Besides, the potential mode of action of the synthesized compounds was determined by employing a molecular-docking study against penicillin-binding protein implicated in anti-bacterial action. Compound M21 was one of the most promising anti-bacterial agents with potential binding affinity against the penicillin-binding protein. This study shed light on novel compounds for further antimicrobial drug development.
